Термин Р. в основном используется для определения процесса синтеза новой нити ДНК на матричной нити ДНК (см. ДНК-зависимая ДНК-полимераза) с целью точного копирования информации, содержащейся в геноме. Р. ДНК является полуконсервативной. Основные стадии этого процесса включают разделение нитей ДНК с образованием репликативной вилки, связывание ДНК-полимеразы и добавление комплементарных нуклеотидов начиная с 3'-конца. Нить, непрерывно реплицирующаяся (ведущая нить, лидерная нить), должна отделяться от др. (запаздывающей) нити, которая реплицируется прерывисто, короткими кусками (Оказаки фрагменты, см.).
