= позиционное картированиеодна из стратегий идентификации и клонирования гена, ответственного за определенное заболевание, при отсутствии данных о продукте этого гена и каких-либо иных генов-кандидатов. При П.к. сначала по результатам анализа сцепления гена с ранее локализоваными генетическими полиморфными маркерами определяют хромосомную локализацию генетического локуса, ассоциированного с заболеванием; затем получают геномные клоны, охватывающие картированный локус (используя соответствующие маркеры, в частности, микросателлитные ДНК), идентифицируют и анализируют присутствующие в них экзоны. Используя целый ряд методов (идентификация GC-островков и экзонов, секвенирование, компьютерный анализ и т.д.), определяют в конечном итоге, какой именно ген ответственен за данное заболевание. П.к. гена, определяющего наследственное заболевание, может быть значительно ускорено при наличии у какого-либо больного цитогенетически видимой структурной перестройки в области локализации этого гена, чаще всего делеции или транслокации; т. обр. были идентифицированы гены хронического грануломатоза, миопатии Дюшенна, болезни Альцгеймера, ретинобластомы, X-сцепленной глухоты, нейрофиброматоза I и ряда других заболеваний.Толковый биотехнологический словарь.
