= неправильное сворачивание[англ. fold — складывать]пространственное сворачивание белка (см. фолдинг), которое отличается от природной, в результате чего белок не только не выполняет свои функции, но может стать причиной возникновения патологий, в частности нейродегенеративных заболеваний; напр., при болезни Альцгеймера происходит образование амилоидных отложений в результате Н.ф. амилоидов: неполностью свернутые молекулы сцепляются друг с другом и образуют нерастворимые волокнистые агрегаты. Н.с. может происходить под воздействием различных факторов (напр., высоких температур) или в результате мутаций в генах. Н.ф. прионов (см. прионы) приводит к устойчивости этих белков к протеазам и способности стимулировать такой же Н.ф. других прионов, что служит основой прионных болезней. Около половины экспрессируемых в E. coli белков, имеющих Н.ф. или находящихся в частично нативном состоянии, образуют комплекс с молекулярными шаперонами (см. шапероны). Впервые Н.ф. обнаружил М. Гольдберг в 1974 г. при изучении восстановления исходной структуры химотрипсиногена (неактивного предшественника фермента химотрипсина) после его денатурации.Толковый биотехнологический словарь.
