Метод приготовления материала для электронной микроскопии, включает замораживание свежей или фиксированной ткани во фреоне или жидком азоте и изготовление срезов в условиях вакуума, при котором происходит частичная сублимация воды из ткани и на ее поверхности проступает рельеф, с такой травленной поверхности получают отпечатки и исследуют их под электронным микроскопом; метод позволяет получать трехмерное изображние объекта.

Процесс обработки микроорганизмов, предназначенных для длительного хранения или электронно-микроскопического анализа.
Широко распространённая процедура в лабораторной практике, в частности для дезинтеграции клеток.
Метод подготовки образца для электронной микроскопии: замороженный образец раскалывают в вакууме, при этом обнажаются мембраны и органеллы, которые затем оттеняются металлом; что позволяет изучить их поверхностную структуру. Полученное изображение очень близко к истинному, так как в данном случае образец не обезвоживается и не подвергается химической фиксации.
Метод, используемый для изучения топографии поверхности, обнажаемой при сколе или срезе глубоко замороженной клетки.
