см. in vitro transcription* * *In vitro транскрипция — метод транскрипции клонированных генов в соответствующий транскрипт in vitro, для чего используется специально приготовленный клеточный экстракт (напр., из клеток HeLa или эмбрионов дрозофилы) и специфический вектор транскрипции ( экспрессионный вектор, см.). Такой вектор должен содержать промотор для РНК-полимеразы и полилинкер. Любая чужеродная ДНК, инсерцированная (вставленная) в полилинкер, будет транскрибироваться под контролем промотора. Наработанный транскрипт может быть использован как зонд при Саузерн- или Нозерн-блоттинге и при гибридизации in situ (см. In situ гибридизация). Для этого иРНК смешивается с клеточным экстрактом из E. coli (для прокариотной иРНК) или с лизатом ретикулоцитов кролика, зародышей пшеницы (для эукариотной иРНК). Такой экстракт практически лишен эндогенной РНК, но в нем есть рибосомные субъединицы (см. Рибосома), все тРНК, аминоацил-тРНК-синтетазы, белки инициации, транслокации, элонгации, трансляционные факторы, аминокислоты, источники энергии (АТФ, ГТФ и др.). Если в экстракт добавлять радиоактивно меченные аминокислоты, то образующиеся полипептиды будут радиоактивными и легко обнаруживаемыми при электрофорезе в полиакриламидном геле и флюорографии.Англо-русский толковый словарь генетических терминов > cell-free translation
