Цинковый палецОдна из основных групп белков, связывающих ДНК. Являются регуляторами транскрипции, содержат характерный домен, который включает 2 цистеиновых и 2 гистидиновых остатков. Эти аминокислоты взаимодействуют с ионом цинка, а расположенная между ними полипептидная цепочка образует петлю в виде «пальца». Цинковые пальцы C2H2 образуют важное семейство связывающихся с ДНК белковых доменов, встречающихся в транскрипционных факторах эукариот. С2Н2-домены, относятся к одному из наиболее перспективных с точки зрения практического применения ДНК- узнающих белков. Предполагается, что изучение этих доменов позволит проектировать белки с цинковыми пальцами, которые будут узнавать специфические последовательности, что актуально в системах диагностики и генной инженерии. С2Н2 домены обязаны своей структурой в первую очередь атому цинка и связывающим его остаткам цистеина и гистидина. Отсутствие атома цинка ведет к распаду структуры.Пространственная структура C2H2 домена. Присутствует одиночный ион цинка, который располагается между бета-шпилькой (т.е., антипараллельным бета-листом из двух бета-тяжей) и альфа-спиралью. Атом цинка координируется двумя цистеинами с одного конца бета-листа и двумя гистидинами с С-концевой части альфа-спирали. На N-концевой части альфа-спирали присутствуют аминокислоты в позициях -1, +2, +3 и +6 (в принятой нумерации номер +1 соответствует началу альфа-спирали), которые отвечают за распознавание и связывание с ДНК. -
