Метод структурного анализа олигонуклеотидов, основанный на том факте, что при добавлении нового нуклеотида к растущей точке транскрипта alpha-фосфат на его 5'-гидроксиле связывается с 3'-гидроксилом предшествующего нуклеотида.
Метод генетической инженерии, основанный на присоединении к 3'-концам цепей, образующих двухцепочечную ДНК, коротких отрезков одноцепочечной ДНК с регулярной последовательностью ( гомополимерных). Ксли каждый подобный гомополимер состоит из нуклеотидов одного вида и если два таких гомополимера с взаимно комплементарными основаниями присоединены соответственно к плазмиде и кольцевой ДНК, то последние, оказавшись рядом, замкнутся друг на друга с образованием рекомбинантной плазмиды.

2 метод обрыва цепи
[греч. methodos — способ познания]метод, применяемый при определении нуклеотидной последовательности ДНК (см. секвенирование ДНК), который основан на полимеразной цепной реакции (см. полимеразная цепная реакция). В качестве матрицы в этой реакции используется одноцепочечная ДНК, нуклеотидную последовательность которой надо определить. Первоначально матрицу гибридизуют с коротким праймером, связывающимся с ее 3'-концом. Затем в реакционную смесь добавляют четыре дезоксирибонуклеозидтрифосфата (дНТФ) и один из четырех дидезоксирибонуклеозидтрифосфатов (ддНТФ). Остановка синтеза новой цепи ДНК-полимеразой (обрыв цепи) будет происходить всякий раз, когда вместо дНТФ в растущую цепь ДНК будет встраиваться соответствующий ему ддНТФ. Для определения последовательности нуклеотидов необходимо осуществить четыре отдельные реакции в присутствии каждого из четырех ддНТФ. Затем четыре пробы вносят в соседние лунки пластины геля и проводят гель-электрофорез, в котором длина пробега каждого компонента обратно пропорциональна длине цепи. Как только фрагменты ДНК визуализированы, нуклеотидную последовательность можно прочесть прямо в геле снизу по направлению к старту в соответствии с очередностью, в которой фрагменты располагаются на отдельных "дорожках". Метод определения нуклеотидной последовательности, основанный на синтезе радиоактивно меченой комплементарной цепи ДНК с использованием в качестве матрицы нативной ДНК. В реакционной смеси присутствуют четыре немеченых дезоксирибонуклеозидфосфата, из которых строится комплементарная цепь ДНК, и модифицированная форма одного из оснований, при включении которого рост цепи прекращается. В результате синтезируются цепочки ДНК различной длины. Цепи разделяют с помощью гель-электрофореза и проводят радиоавтографию. Последовательность оснований считывают непосредственно с фотоплёнки.

Метод определения нуклеотидной последовательности, при котором метят 3'-конец молекулы ДНК радиоактивным фосфатом (32PO43–). Затем ДНК обрабатывают реагентами, расщепляющими эту цепь около определённых оснований. С помощью электрофореза в полиакриламидном геле разделяют фрагменты разной длины. Последовательность оснований считывают с плёнки, полученной методом радиоавтографии.

Метод определения последовательности оснований в самой РНК, для чего сначала метят 5'- конец или 3'-конец молекулы РНК с помощью 32PO43–. Затем меченую цепь расщепляют с помощью ферментов, специфичных к определённым последовательностям. Используя радиографию, получают карту, по которой находят последовательность оснований.
Метод определения последовательности оснований, в котором вначале синтезируют цепочку ДНК, комплементарную РНК, а затем определяют её нуклеотидную последовательность. Далее с помощью метода химического расщепления для секвенирования ДНК получают радиоавтограмму.
