1 DNA Probes
ДНК-ЗондыЗонд для нуклеиновой кислоты – короткоцепочечная ДНК, которая обнаруживает и связывает комплементарную последовательность в образцах, содержащих одноцепочечные фрагменты ДНК или РНК, что позволяет определять целевые последовательности. Анализ с использованием зондов основан на важнейшей реакции гибридизации, протекающей спонтанно между двумя комплементарными цепями ДНК/ДНК или ДНК/РНК. Как и в иммуннохимическом анализе, для обнаружения гибрида необходимо, чтобы зонд содержал метку. Для обнаружения гибрида были разработаны различные прямые и косвенные методы. Прямая маркировка предполагает непосредственное присоединение метки к последовательности зонда. Косвенное мечение - связывание антитела с гибридом ДНК/ДНК или ДНК/РНК. Как и в иммунохимическом анализе, более предпочтительны зонды, где метка не является радиоактивной, главным образом ввиду радиационной опасности, сложностей утилизации и небольшого срока хранения реагента последних. Кроме того, факторы, влияющие на предел обнаружения гибридизации проб, содержащих меченые зонды, схожи с факторами, характерными для иммунохимического анализа. Поэтому разработка простой, недорогой и чувствительной системы прямого обнаружения, не связанной с мечением, очень желательна. -
