1) бактериальный фермент, катализирующий удаление 5'-концевой фосфатной группы с концов молекул ДНК или РНК при высоком рН. Используется в генной инженерии (см. генетическая инженерия) для предотвращения закольцовывания и димеризации плазмидных векторов ДНК после их разрезания рестриктазами (см. рестриктазы). Обработка ДНК Щ.ф. увеличивает вероятность получения рекомбинантных молекул, так как образование кольцевых молекул в этом случае может происходить только при наличии чужеродной ДНК (вставки), у которой имеются 5'-концевые фосфаты на обоих концах. Щ.ф. также используется в иммуноферментном анализе в качестве маркера, в процессе мечения нуклеиновых кислот биотином или диоксигенином;2) фермент, образующийся в различных органах высших организмов (кости, печень, почки и др.), содержание которого увеличивается в моче или крови при некоторых типах заболеваний, используется как диагностический показатель.Толковый биотехнологический словарь.
